According to Wikipedia,
Focus stacking is a digital image processing technique which combines multiple images taken at different focus distances to give a resulting image with a greater depth of field (DOF) than any of the individual source images.

In macrophotography and in photomicrography, by closing the aperture at F/16 - sometimes even beyond F/8 - the depth of field increases, but phenomena of diffraction appear and the image becomes increasingly blurred and less distinct.

With focus stacking you can combine in one resulting image the sharpest areas of different images taken on the same subject at different focus distances and using the sharpest aperture of the lens.

Here you see the stack of 5 images of the same subject (velvet-ant wasp, Hymenoptera Mutillidae) taken at different focus distances using f/4 aperture and the resulting image of the focus stacking procedure:
